I guess it depends on how far you'll stretch what's been done before but if you look at Hendrix, he was amazing and did things with a guitar that definitely had not been done before. There are others but that's the one that stands out most in my mind. Often times what occurs is that someone will be the first to bring a particular sound to the mainstream. If I were to guess where the next sound that will change music will come from, I would say that it will have to do with creating rhythms without drumbeats. I know this has occurred before (my personal favorite being John Mayall's 'Turning Point') but I think someone's going to come up with something new in that area. But music is similar to technology where most innovations are gradual with the occasional breakthroughs which are usually putting together existing technology in a new way but, every so often, some major discoveries do occur.
